ABSTRACT

In order to ensure the safety of the vaccine cold chain to the greatest extent, the newly regulations still require daily manual recording of temperature data and operation conditions based on the configuration of the automatic temperature measurement and alarm system. Therefore, on the basis of Shanghai Vaccine Cold-chain Monitoring System, Shanghai has introduced an electronic signature technology to implement a new model of remotely checking the temperature by mobile-phone and keeping records of the electronic signature as certificates. This technology reduces the on-site workload of cold chain management personnel. It also guarantees the authority, legitimacy, and non-tampering of recorded information through blockchain certificate storage technology. The application of the electronic signature technology is compatible with technical defense and civil defense methods, and is worthy of further promotion and use.

ABSTRACT

OBJECTIVE@#To explore the influencing factors and countermeasures of infection in leukemia patients after allogeneic peripheral blood hematopoietic stem cell transplantation.@*METHODS@#A total of 126 patients with leukemia admitted in our hospital from August 2016 to March 2018 were selected. The number of infected patients after transplantation was recorded, and the causes of infection were analyzed.@*RESULTS@#Among the 126 patients, 43 were positive for infection, and the infection rate was 34.13%. A total of 89 pathogens were detected, of which bacteria accounted for 64.05%; virus accounted for 22.47%, and fungi accounted for 13.48%. The patient's age, donor type, pre-transplant infection, prophylactic use of antibiotics and aGVHD all were factors influencing the patient's infection (P<0.05). The follow-up results showed that the incidence of infection in the intervention group significantly decreased after intervention with prevention program (P<0.05). After reasonable nursing intervention, the incidence of infection in the intervention group after follow-up for 12 months was lower than that in the control group (P<0.05).@*CONCLUSION@#Pre-transplant infection and prophylactic use of antibiotics are factors influencing the infection after allogeneic hematopoietic stem cell transplantation. The incidence of infection can be reduced by reasonable infection prevention.

ABSTRACT

Objective To evaluate the efficiency of REPLI-g® Single Cell Kit for sample DNA amplification, and explore its application value in forensic trace DNA amplification. Methods Three DNA extraction kits were selected to extract DNA from peripheral blood of 10 unrelated individuals. The DNA yield and purity of the three DNA extraction kits were compared. According to the results of comparison, one DNA sample was selected to concentrate and dilute, then used as the initial sample of whole genome amplification (WGA). REPLI-g® Single Cell Kit was used to amplify the initial sample at the whole genome level. The amplification yield and amplification times were calculated, and the distribution of DNA fragments was detected by agarose gel electrophoresis. Goldeneye® DNA ID System 20A Kit was used to perform the STR typing of the initial sample and DNA samples amplified at the whole genome level to evaluate the performance of REPLI-g® Single Cell Kit in trace DNA amplication in terms of purity and yield as well as the success rate of STR typing. Results After comparison, one DNA sample was selected from QIAsymphony® DNA Investigator® Kit extracts to concentrate and dilute as the initial sample of WGA. After amplifying the whole genome of a series of initial samples by REPLI-g® Single Cell Kit, the lowest average of amplification yield reached 8.77×103 ng, while the average of the corresponding amplification times reached 1.40×106. DNA fragments were large and concentrated. The STR typing success rate of WGA samples became lower with the decrease of initial samples used, but when the amount of samples was lower than 0.5 ng, the STR typing success rate of samples after DNA WGA was higher than that of samples without DNA WGA. Conclusion REPLI-g® Single Cell Kit can increase the yield of template DNA. Especially for trace DNA, the STR typing success rate can be improved to a certain extent.

ABSTRACT

To study the effect of recombinant human growth hormone [rhGH] therapy on carbohydrate, lipid and protein metabolisms of Turner syndrome [TS]. Total 45 patients with TS admitted between Jul. 2008 and Jun. 2011 were involved in this study. All patients received the clinical evaluation of body fat, plasma lipids, proteins and oral glucose tolerance test [OGTT] before and after rhGH therapy. Our results indicated a significant decrease of body fat [FAT%] from 23.56 +/- 4.21 to 18.71 +/- 2.23 but no obvious change on the level of fat mass [FM] [p > 0.05] was observed after rhGH therapy. We also detected significant changes on plasma high-density lipoprotein cholesterol [HDL-C] from [1.65 +/- 0.58 mmol/L] to [2.20 +/- 0.65 mmol/L] and low-density lipoprotein cholesterol [LDH-C] from [2.55 +/- 0.55 mmol/L] to [2.10 +/- 0.54 mmol/L] after rhGH exposure. However, no statistical significance was detected on the level of plasma triglyceride [TG], cholesterol [CHO]. Interestingly, the levels of plasma retinol binding protein [RbP] [32.55 +/- 4.28mg/L], transferrin [TRF] [2.95 +/- 0.40 mg/L], serum albumin [PRE] [250.00 +/- 45.50 mg/L] and albumin [propagated] [33.58 +/- 4.25 mg/L] were significantly increased. When it goes to the oral glucose tolerance test [OGTT] test, there were 10 impaired glucose tolerance [IGT] cases among all patients before and after rhGH therapy. No significant change was observed on homeostasis model assessment- insulin resistance [HOMA-IR] level during rhGH intervention. Abnormal lipid and protein metabolisms of the children with TS can be improved with rhGH therapy for 6 months

ABSTRACT

Objective: By observing the effect of Kanglaite (KLT) Injection in combination with gefitinib on angiogenesis in mice with Lewis lung cancer, to explore its possible mechanism. Methods: Lewis lung cancer model was established in C57BL/6 mice. Forty model mice were randomized into 4 groups: control group, KLT treatment group, gefitinib treatment group, and KLT+gefitinib treatment group. Using immunohistochemistry and RT-PCR technology to observe and compare tumor growth and angiogenesis in each treatment group. Results: The average weight of the tumor in KLT+gefitinib treatment group was lower than those in the KLT treatment group and the gefitinib treatment group; the difference was statistically significant (P < 0.01). Judging from the inhibition rate, KLT, gefitinib, and KLT+gefitinib can inhibit the growth of the transplanted tumor, especially for the combination treatment. The microvessel density (MVD)-positive labeling index (LI) in control group, KLT treatment group, gefitinib treatment group and the KLT+gefitinib treatment group were 24.35±1.06, 18.25±1.36, 20.09±1.46 and 14.46±0.98, respectively. The MVD of the KLT+gefitinib treatment group was lower than those of the two single drug groups; the difference was statistically significant (P < 0.01). The expression levels of VEGF and KDR proteins in the KLT treatment group and the gefitinib treatment group were lower than that of the control group (P <0.05) and higher than that of the KLT+gefitinib group (P < 0.05). The expression levels of VEGF and KDR mRNAs in the KLT+gefitinib group was lower than those of the two single drug groups; the difference was statistically significant (P < 0.05). Conclusion: KLT combined with gefitinib treatment may exert synergistic effect on anti-angiogenesis, so as to obtain synergistic anti-tumor effect. Copyright © 2013 by TUMOR.

ABSTRACT

Objective To study the relationship of HPV pseudo-neutralizing titers detected by two different reporter genes: Zoanthus sp. green fluorescent protein (ZsGreen) and secreted alkaline phosphatase (SEAP) , and the relationship between HPV the pseudovirus-neutralizing antibody titer and the antibody titer determined by ELISA method. Methods The plasmids with expression cassettes of the HPV capsid protein L1 and L2 genes after codon optimization and the plasmid with reporter gene (ZsGreen or SEAP) were co-transfected into 293FT cells. The cell lysate supernatants were collected after 48 h culture, then the pseudovirus was purified through POROS column chromatography from the supernatants. After the titer of pseudovirus bulk were measured, HPV-16 and HPV-18 pseudovirus-neutralization assays were carried out for determining the titer of sera collected from immunized mice with HPV candidate vaccine and Gardasil HPV vaccine. Results In statistical analysis, the two reporter gene systems for the detection of the pseudovirus neutralizing antibody titer are highly relevant to each other (Spearman coefficient; r = 0. 760). And their neutralizing antibody titers bear a high degree of correlation with the antibody titer (Spearman coefficient: r= 0.577 and r =0. 741). Conclusion ZsGreen and SEAP pseudovirus neutralizing antibody titers are highly relevant to each other. The neutralizing antibody and the antibody titer are also relevant. These results reveal some mechanism of HPV vaccines to prevent the virus from invading the host cells, and are absolutely useful in the protection efficiency evaluation of the HPV-16 and HPV-18 candidate vaccines.

ABSTRACT

Objective To understand health seeking behavior and its influential factors to reproductive tract infections (RTIs) on women at reproductive age in the rural areas. Methods 54 540 fertile women aged 15-49 were surveyed by a stratified-cluster-random sampling method and gynecological examination were conducted in two steps: converging at the clinics, and then visiting their households, later, 31 624 women who had at least one RTI symptom were chosen. Results Among all the women at reproductive age, the rate of having at least one RTI symptom was 59.8% with the means of RTI symptom as 1.66±0.89. 15 989 women went to see doctors out of the 31 624 women who had RTI symptoms, with a proportion of 50.6 %. The results of logistic regressy showed that those women whose husbands having higher education level, higher income, more RTI symptoms and better knowledge on RTI were more easily to go to the hospitals. However, those women whose husbands working out of the county, having older first bearing age and more numbers of pregnancy were less likely to go to the hospitals. Reasons that refrained them from going to see a doctor would include: 2137(13.7%) did not know that RTI was a disease; 7443(47.6%) of them thought that every woman were bound to have at least one symptom and it did not matter; 1629 (10.4%) of them felt shameful; 349 (2.2%) learned that the diseases were incurable; 975 (6.2 % ) felt the cost of treatment was too expensive; 2101 (13.4 %) had no time; 1001 (6.4 %) would treat themselves through buying medicines over the counter. Conclusion RTI symptoms were quite prevalent among women at reproductive age but the rate of seeing a doctor was low and caused by multi-factors. Health education and gynecological census in increasing the curable rate of RTIs should to be strengthened.

ABSTRACT

OBJECTIVE To study the isolation and resistance tendency of Acinetobacter calcoaceticus-baumannii to antimicrobial agents from 1998 to 2004 to provide valuable data for infection prevention and therapy. METHODS We reviewed the isolation rates, distribution in clinical specimens and wards, and the resistance rates of(A.calcoaceticus-baumannii)to 14 kinds of antimicrobial agents from 1998 to 2004. RESULTS There was an increasing tendency of isolation rates of A.calcoaceticus-baumannii every year, which was 0.18% in 1998 but 1.48% in 2004.In the seven years, there was the highest isolation rate of 70.58% in specimens from respiratory tract, the next was from the urine(9.42%),and blood(4.63%).Concerning the wards distribution,ICU had the highest rate of 47.28%.In 1998,A.calcoaceticus-baumannii had resistance rates more than 50% only to one kind of antimicrobial agents(aztreonam),but in 2004,it had increased to thirteen kinds(except cefoperazone/sulbactam).About the fourteen kinds of antimicrobial agents we inspected, that were increased in their resistance rate. The highest increasing of resistance rate was ceftazidime from 11.1% in 1998 to 88.9% in 2004,the imipenem was second for 0.0% to 64.8%,and the third was sulfamethoxazole/trimethoprim form 0.0% to 64.0%,while there still was an increasing resistance tendency to them. CONCLUSIONS The clinical isolation rate of A.calcoaceticus-baumannii is increasing, and it has higher resistance rates to many antimicrobial agents as well as an increasing resistance tendency to relatively susceptive antimicrobial agents every year.So physicians should prescribe on the basis of antimicrobial agents susceptibility tests in vitro.
